The T-Pro BCA Protein Assay Kit is a bicinchoninic acid-based detergent compatible formulation for colourimetric detection and quantification of total protein. This method was based on the reduction of Cu + 2 to Cu + 1 by protein in an alkaline medium with highly sensitive and selective colourimetric detection.
The purple reaction product from this assay forms and exhibits a strong absorbance at 562 nm that is nearly linear with increasing protein concentrations over a wide working range (20-2,000 µg / ml). The reaction that occurred in the T-Pro BCA Protein Assay Kit is not a true end-point method; that is, the final colour continues to develop.
However, after incubation, the rate of continuous colour development is slow enough to allow a large number of samples to be analyzed together. Therefore, protein concentrations are generally determined and reported with reference to bovine serum albumin (BSA) standards.
A series of dilutions of known concentration are prepared from the protein and analyzed along with the unknowns before the concentration of each of the unknowns is determined based on the standard curve.
